What is EpiTYPER?
EpiTYPER is a MALDI-TOF mass spectrometry-based bisulfite sequencing method that enables region-specific DNA methylation analysis in a quantitative and high-throughput fashion. The technology interrogates 10s-100s of samples and CpG sites in amplicons from 200 – 600 bp and detects down to 5% differences in methylation.
How do you test for DNA methylation?
Currently, there are three primary methods to identify and quantify DNA methylation. These are: sodium bisulfite conversion and sequencing, differential enzymatic cleavage of DNA, and affinity capture of methylated DNA (1). Restriction enzyme based differential cleavage of methylated DNA is locus-specific.
What is an epic array?
The Infinium methylation EPIC array allows the interrogation of methylation patterns at the genome-wide level, covering more than 850,000 methylation sites. This array has shown to be very popular and is an ideal solution for epigenome-wide association studies even for FFPE samples.
What is methylation assay?
DNA methylation assays could be considered in routine toxicity screening as they provide comprehensive information on local and global molecular events compared to traditional other genetic assays [212]. From: Environmental Epigenetics in Toxicology and Public Health, 2020.
How does a methylation array work?
Methylation arrays enable quantitative interrogation of selected methylation sites across the genome, offering high-throughput capabilities that minimize the cost per sample. Array-based methylation studies can provide valuable insights into the regulation of gene expression.
Why is it important to study DNA methylation?
DNA methylation is essential for silencing retroviral elements, regulating tissue-specific gene expression, genomic imprinting, and X chromosome inactivation. Importantly, DNA methylation in different genomic regions may exert different influences on gene activities based on the underlying genetic sequence.
What diseases does microarray test for?
Chromosomal microarray (CMA) is increasingly utilized for genetic testing of individuals with unexplained developmental delay/intellectual disability (DD/ID), autism spectrum disorders (ASD), or multiple congenital anomalies (MCA).
What is epidesigner for DNA methylation?
STEP 1: Assay Design EpiDesigner is an online automated design tool for DNA methylation experiments on the MassARRAY System. Just enter your target sequences and the software determines primer designs for the most complete DNA coverage.
What is Step 3 of the epityper assay?
STEP 3: PCR, In vitro transcription, and RNA Cleavage Design The EpiTYPER Assay starts with PCR using T7-promoter-tagged reverse primers to amplify the target regions while preserving the bisulfite-induced sequence changes.
What is epityper MassARRAY?
The DNA methylation analysis technology, EpiTYPER MassARRAY, provided by Agena Bioscience (previously Sequenom Inc.), is one of the most reliable quantitative methods available today for DNA methylation analysis.
How to distinguish between methylated and non-methylated cytosine residues?
The methylated and non-methylated cytosine residues in the original genomic DNA are easily distinguished using EpiTYPER Software. STEP 1: Assay Design EpiDesigner is an online automated design tool for DNA methylation experiments on the MassARRAY System.
